
Dermatan sulfate is a natural glycosaminoglycan found mostly in skin, but also in blood vessels, heart valves, tendons,lungs and intestinal mucosa. In addition to its role as a major constituent of the skin and other organs, Dermatan Sulfate is believed to play a part in repairing wounds, regulating the coagulation of blood, and responding to infections, though its role in these processes is not well understood.
Due to its important role in cell growth, differentiation, morphogenesis and cell migration this substance is mainly used for cosmetic purposes. However, it’s clinical potential is believed to be very significant. Dermatan Sulfate performs interesting biological activities, which should help in the development of Dermatan Sulfate-based therapeutics, such as drugs for parasitic and viral infections, regenerative medicine, anti-tumor drugs, or simply as a marker of significant disease progression.
Biological activities of Dermatan Sulfate chains are likely to involve various growth factors, and specific Dermatan Sulfate chains having distinctive structures and properties are able to recruit factors and/or potentiate their activities, suggesting that minute amounts of functional Dermatan Sulfate chains can be utilized as active biomolecules. To this aim, new bioactive sources of Dermatan Sulfate may represent potential drugs for future research and development, as well as for a better understanding of the structure–function relationship, and would enable the production of this polysaccharide with its distinctive composition, structure and biological activities.


Dermatan Sulfate, also known as Chondroitin Sulfate B, is found in a wide variety of tissues in virtually all animals and is produced by extraction and purification. Dermatan Sulfate is a linear polysaccharide assembled as disaccharide units containing GalNAc or IdoA joined by β-1,4 or 1,3 linkages, respectively.


Anticoagulant and antithrombotic activities
A well-studied Dermatan Sulfate binding interaction occurs with heparin cofactor II that acts by inhibiting the procoagulative activity of thrombin and this effect is enhanced 1000-fold in the presence of Dermatan Sulfate.
Dermatan Sulfate in wound repair
As is well known, Dermatan Sulfate is a major constituent of the skin and, considering its location and activity in coagulation and cell growth, and its capacity to interact with several proteins, the function of Dermatan Sulfate in wound repair is an active area of investigation.
Dermatan Sulfate for infections
Dermatan Sulfate (and related Proteoglycans) have been associated with the capacity to modify resistance to infectious disease.
Dermatan Sulfate in the central nervous system
The Central Nervous System is a rich and complex source of Proteoglycans composed of Dermatan Sulfate/Chondroitin Sulfate chains. Particularly high levels of Chondroitin Sulfate/Dermatan Sulfate proteoglycans have been observed during brain development having the capacity to finely tune expression patterns suggesting important functions at all stages of development.
Dermatan Sulfate as a potential anti-tumor drug A clear correlation between the accumulation of Chondroitin Sulfate/Dermatan Sulfate proteoglycans and cancer progression has been demonstrated. Dermatan Sulfate interacts with many growth factors and cytokines that are involved in cancer and cancer formation. As consequence, fine structural alterations of Dermatan sulfate chains and its properties can influence the biology of cancer cells, contributing to cell growth and migration. The specific sequences that bind to several key proteins are yet to be identified but its investigation and characterization may lead to molecular targets for cancer diagnostics and therapeutics.
